Assays that can detect immunogenic responses to a biotherapeutic drug, indicated by the presence of anti-drug antibody (ADA), are required in preclinical and clinical studies through to post-market surveillance. Such assays must be drug tolerant i.e. able to detect ADAs at low concentrations in the presence of free drug.

Established sample pretreatment steps to improve drug tolerance are integrated into the automated, nanoliter-scale immunoassay workflow of a Gyrolab ADA CD. A Gyrolab assay protocol and Rexxip ADA buffer facilitate assay development. Dedicated Gyrolab ADA Software guides the user through a standard ADA analysis workflow from development and validation to screening and confirmatory analysis.
IMPORTANT NOTE for Gyrolab workstations users
Running Gyrolab ADA CDs requires installation of the Gyrolab ADA Software module with Gyrolab Control v 5.4 or higher and Gyrolab Evaluator v 3.3 or higher.
Gyrolab workstations delivered after October 1, 2011 will be installed with Gyrolab Control v 5.4 or higher and Gyrolab Evaluator v 3.3 or higher.

Using Gyrolab Bioaffy CDs
As seen in earlier presentations and publications, Gyrolab workstation users have used Bioaffy CDs to successfully develop ADA assays and benefit from working at nanoliter scale. Sample pretreatment, when needed to improve drug tolerance, has been performed outside of the Bioaffy CD. Assays requiring sample pretreatment can now be transferred to the time-efficient, drug-tolerant Gyrolab ADA solution.
